Use of X-ray Microscopy for Confirmation of Crystallinity Detection in Amorphous Formulations by Electrospray Laser Desorption Ionization Mass Spectrometry Imaging

The use of mass spectrometry imaging for crystallinity detection offers improved matrix selectivity and sensitivity over the techniques, such as X-ray microscopy Raman spectrometry, that are traditionally used with this work. Crystallinity is observed in electrospray laser desorption ionization (ELDI-MSI) a high-intensity agglomeration analyte spatially resolved image. As an indirect method detection, confirmation method’s ability to detect crystallization amorphous formulations needed by directly correlating observations tablet ELDI-MSI those established technique. Micro-computed tomography (micro-CT) has necessary investigation ideal evaluating correlation ELDI-MSI. In work, micro-CT followed ELDI-MSI, same location on tablets miconazole spiked trace levels crystalline miconazole, were analyzed. Crystals detected matched crystals but chemically unidentified micro-CT. results study conclusion about effectiveness complimentary technique indirectly enabling formations API presented.
